Though I really like my XDs and Glocks. If I had to have only one of my guns to make it though, I would probably choose my Sig P220 Stainless. It is dependable, accurate, and durable and can handle any ammo I have tried to put through it without a hitch. It is a little heavy to carry, but if I had to, I know I could.
I have thouhgt about this question a lot, not to mention having owned and tried out many handguns. Currently, I'd have to go with the Glock 19. It is midsized, big enough to shoot a lot and small enough to carry. It has great mag. capacity and fires a round that should be available in emergency situations. It's reliable, tough and won't rust. There are enough around to find spare parts easily.
I love my G23 but may go to something not as thick someday.

I'd go with the Glock 19 as well, although I find myself carrying it less and less often. It may be the best "compromise" pistol out there for defensive purposes.
Pick one up and you will never want to put it down!!!!!
I carry a glock26, but I would go with a glock19 if I could only have 1 for everything
Only one for everything then I would have to go with either a model 29 S&W or a 629 S&W. 4" barrel
Then I can use either 44Mag or 44Special. They can be loaded for Snakes(shot shell), Protection, Hunting (from squirrels to Bear and everything in between).
Little harder to conceal then my 1911 but not much, Just need the right Holster.
If I only get one hand gun: I'd go with my Para ord 1911A1 SSP, with all the MIM parts replaced with milled versions (I'm working on doing that now).
I'd also buy a rifle to keep next to it alot sooner (sence I cant spend that money on hand guns:).
+1 for CC. I wouldn't trade mine. However, I have found that I would like to have the 21 for duty use. If I could have only one, I might opt for the full size and deal with CC issues.

Glock 23. 13 rds. in the mag, decent "man stopping" caliber, not too big not too small. Controllable recoil. Endless supply of gear designed for it. Swap barrels for .357 sig. Lots of police carry it, so it might slow them to shoot me if I have a bad guy at gunpoint when they roll up. Very versatile. Easy to disassemble and clean, easy to repair if needed. Night sights and a good holster and belt will do you fine.
