For me it would be a gun I don't currently own. I think it would be a SP101 with a 3" barrel. Small enough for carry duty and still an excellent field gun for outdoor activities....it's on my next purchase list...

    Only one handgun?

Geez, I have too many uses for just one. If I had to pick just one to do everything I demand of a handgun it would be a 4" .357 Magnum. Most likely a 686 or a GP100.

It's not so much the gun, but the caliber I love.

If I had to choose one it would be my Glock G29SF. Glock reliability, 10mm firepower with a good variety of loads available, easily concealed, good for HD and hunting. With a simple barrel swap I can also shoot .40, .357SIG and 9x25 Dillon. Throw in some G20 15rd mags and I have plenty of capacity also.

It can basically do everything I'd ever need.

For the time being I'd have to say my G26. It's not the ideal gun for all uses but it does fill the majority of my needs for SD purposes.

I would say my Glock G26 (9MM) also because of all the guns I've owned
its the best compromise I,ve found between power, size, reliability, durability,
    capacity and ease of maintenance.

It would be either my Wilson CQB or my Les Baer SRP. I can dress around the size.

You said one Gun, but you didn't say anything about parts. Therefore, I would choose my Glock 29, with just a few modifications I could shoot in addition to 10mm. 40 S&W, and 357 Sig. Making it the Swiss army knife of handguns.

Glock 19 because it has 15+1, it's reliable, easy to maintain, accurate, easy to conceal, and it's the only handgun I own at this point.

I See we have some Kimber fans. Kimber makes great 1911 pistols. I like the Ultra Carry too. It is my personal feeling that you shoot what you are comfortable with. I have several 9mm pistols that I am confident can do the job most of the time. 9mm usually works fine for FBI though it has proven to be inadequate in some shoot outs with armored assailants however FBI are not issued 45's.
9mm is my choice most of the time. I have a .40 and a .45 ACP and a coupe .22's. I think it is possible to take down a bad guy with .22 LR HP very easily though I have never done so and hope it never happens. I do keep my Kimber and my Sig's loaded and ready all the time. I usually only carry the more concealable Walther PPS and would not try to conceal the big Kimber except in the winter.
I do carry a large LED Mag-lite flashlight in the car and at home. This can be used as a powerful light or club as needed.

I was about to say my Smith & Wesson 686...stainless steel reliability, fairly high capacity for a wheelgun (7), and the fact that it makes it appear that I can actually shoot.

But the edge has to go to my Smith & Wesson 327 - - while I worry about the durability of the frame (what is it? scandium? titanium?), it is lighter, marginally more accurate, holds 1 more round and has a better sighting system (tritium big dot up front vs. an orange insert in a blade).

    It's close. Real close.
