I have attached a blueprint of my bedroom layout and would appreciate any advice on this. I'm trying to have the best tactical advantage in the event of a B&E during the night. I have listed where weapons are located. Couple of things to keep in mind, tv has to stay stationary for the most part, cable coming in from under the house. Also, If I move my bed, I don't want the headboard in front of the bedroom door.

Just change the door out to a solid, 1 hour fire door with a good lock system.
If the BG breaks the window you could wind up with glass in your face.
Decent idea, although an expensive one. Have you ever considered a door alarm? If you are on a budget a cheap one can be had at Walmart. Sometimes ample warning of the BG attempting entry (which will wake you before the threat is standing over your bed) is more helpful in a tactical situation than how your furniture is arranged (unless you have bulletproof furniture). Nothing that I saw listed on that layout would stop, or even slow down a bullet enough to make a difference. Those few seconds could be life or death. I would address that issue first. If doing this, be certain of having a solid door/jam, or a security bar angled into the floor that disallows even heavy kicks. The point is to gain time, so that you can get to that pistol "on the night stand" or the shottie in the safe.
Put the shottie above your bed with a trigger lock on it. Attach the key to the trigger lock to your house keys. When you go to bed at night put the trigger lock key in the lock this way you just turn and the lock will fall to the ground.
arrange a system where you are surrounded by 5 feet of steel on every side, and have a magic code word where when you say it, all your weapons swing down from a hidden latch and into your hands.
Your layout looks pretty good, Sundance suggest you may want a revolver for under your pillow?
Depending on how much you like your wife, have her sleep on the side closest to the door.
Seriously, I have my Glock on the nightstand at the ready. I've thought about fixing a holster for it behind the headboard or something so it wasn't easilly accessable to someone who didn't know it was there.
