Right in the ear works as well. Their skull isn't particularly rugged, but you want to make sure you have a pretty perpendicular shot so the round penetrates and doesn't ricochet off. I usually shoot right in front of the base of the ear (the temple). It's 100% so far.
If you're talking about hunting deer with a handgun, I'd have to go no lighter than a 10mm/.41 magnum, using a good hollow point (Hornady HP/XTP works very well).
