First two rounds are #6 shot small game load, followed by three Federal low recoil 9 pellet 00 buck, with five more on the stock.

I'd be concerned about having the time to get to the third shot, if the chips were down and time was tight. It's not like the time clock is of our own making, in such a situation.

#1 BUCKshot seems to give the best blend of penetration and number of pellets. A shell of #1 buckshot would cause more trauma than 00 buckshot. Buckshot also penetrates fewer walls than most typical handgun and rifle ammo. See testing at The Box O'Truth.

BIRDshot of any variety is for little birds and small game. It will not reliably penetrate to the vitals on a human. If you use birdshot for defense, you should reconsider, IMHO.

Slugs are good if you are in a rural area and may need a longer range shot outside the home. Inside the home, they have a significantly greater risk of overpenetration, compared to buckshot. They also give away the major advantages of the shotgun - the simultaneous delivery of multiple wounds, and the greater ability to hit a moving target. If I felt a need to use slugs outside, I would grab my Mini 14 instead.
